Leica STED Confocal, Super-Resolution, and FLIM

Confocal fluorescence microscopes can generate high-resolution three-dimensional images of thick specimens. With a 63x/1.4 oil lens you can get a resolution of about 0.2 microns laterally and about 0.5 microns in depth. Our Leica STED microscope, can be used in regular confocal mode, but it also has a super resolution STED (Stimulated Emission Depletion) mode that allows for higher resolution imaging down to ~50nm. It is equipped with 405nm and 440nm solid state confocal lasers, a white light laser (470-670nm) and STED lasers 592, 660 and 775nm. There are two regular PMT detectors and 3 higher-sensitivity HyD detectors, as well as spectral detection with a resolution of down to 1nm. It has an inverted microscope base and a stage top incubator allowing imaging of live cells in coverglass-bottom petri dishes or chambers. The Adaptive Focus Control (AFC) keeps the focus constant during long live cell experiments. This microscope can also be used for FLIM (Fluorescence Lifetime Imaging) and FCS (Fluorescence Correlation Spectroscopy). Optional post processing with Hyvolution software allows for improved confocal resolution.
